Increased Expression of AQP7 in the Skeletal Muscles of Diet Induced Obesity Mice | Chapter 12 | Highlights on Medicine and Medical Research Vol. 8

 Aquaporins 7 and 9 are membrane proteins that belong to the aquaglyceroporin family, which transports glycerol as well as water molecules. Glycerol is a direct source of glycerol-3 phosphate, which is needed for the synthesis of triglyceride, the main component of lipid droplets found in skeletal myofibers. We hypothesised that the expression of AQP7 and AQP9 in skeletal myofibers of diet-induced obese (DIO) mice will be different from that of control chaw-fed mice. The levels of AQP7 and AQP9 RNA and protein in the quadriceps femoris muscles of DIO mice and regular control mice were investigated. Mouse AQP7 mRNA levels in skeletal muscles were significantly higher in DIO mice compared to normal control mice (P 0.01), but mouse AQP9 mRNA levels were not different between the two classes (P > 0.05), according to real-time quantitative RT-PCR analysis. In oil red O stain samples, the myofibers of DIO mice contained numerous lipid droplets. Immunohistochemistry of DIO mouse muscles revealed increased expression of AQP7 at myofiber surface membranes, while AQP9 expression was comparable to that of normal control mice. These results suggest that increased AQP7 expression in DIO mouse muscles promotes glycerol secretion from myocytes.
