The oral cavity has a diverse population of mesenchymal stromal cells (MSCs). Dental-derived MSCs-based approaches have shown significant potential among regeneration therapies. Dental-derived MSCs have been used in several preclinical investigations and clinical trials for the treatment of dental and non-dental diseases including as neurodegenerative diseases, autoimmune diseases, and orthopaedic problems. Dental-derived MSCs, on the other hand, vary in their origin tissues, individual donors, clonal subpopulations, and single cells. Variation in regeneration potential across cells in dental-derived MSCs cultures could skew trial outcomes and stymie or halt the adoption of dental-derived MSCs therapy in clinical practise. Although variation is unavoidable, our limited ability to detect and manage heterogeneity makes manufacturing dental-derived MSCs therapies difficult. We initially explored the heterogeneity of dental-derived MSCs before moving on to the important markers and epigenetic alterations associated with dental-derived MSC proliferation, differentiation, immunomodulation, and ageing. These messages aid in the regulation of the composition and function of dental-derived MSCs, allowing cell therapy to be more quickly implemented in clinical practise.
