An Advanced Study on Development and Validation of RP-HPLC Method for Simultaneous Estimation of Mesalamine and Prednisolone in Bulk and Formulation | Chapter 1 | Technological Innovation in Pharmaceutical Research Vol. 9

 A simple, precise, accurate, sensitive, and economical reversed phase high performance liquid chromatographic method for the simultaneous determination of Mesalamine (MSL) and Prednisolone (PRD) in combined dosage forms was developed and validated in accordance with the analytical parameter mentioned in the ICH guidelines. The drugs were separated using chromatography on a C-18 Phenomenex column (250 mm 4.6 mm i.d.) with an acetonitrile:phosphate buffer (20:80 v/v) mobile phase (pH adjusted to 6 with Orthophosphoric acid). The detection was carried out in a 330 nm is the wavelength. The retention times for Mesalamine and Prednisolone were 4.373 minutes and 1.589 minutes, respectively. Meselamine was found to be linear in the range of 5-25 g/ml, and Prednisolone was found to be linear in the range of 2-10 g/ml. Mesalamine and Prednisolone were shown to have recovery rates of 99.57 percent and 100.63 percent, respectively. Mesalamine has a correlation coefficient of 0.9973, whereas Prednisolone has a correlation coefficient of 0.9984. With a smaller RSD, the approach demonstrated appropriate precision (less than 1 percent ).
