Evaluation of Methanolic Leaf Extract of Parkia biglobosa (African locust bean, Jacg, Benth) Leaves against Trypanosoma evansi | Chapter 15 | Technological Innovation in Pharmaceutical Research Vol. 10

 Parkia biglobosa leaves were extracted with methanolic solvent at concentrations (250-1000 g ml-1) in our ongoing investigation to evaluate the presence of antitrypanosomal compound(s) from medicinal plant. Methanolic plant extract (MPE) was investigated for trypanocidal efficacy against Trypanosoma evansi. At the right conditions, Vero cells were cultured in Dulbecco's Modified Eagle Medium (DMEM) and supplemented with 20-40% foetal calf serum (FCS). On Vero cells without FCS, an in vitro cytotoxicity test of P. biglobosa methanolic leaf extract at doses (1.56-100 g ml-1) was performed. The effects of immobilisation on trypanocidal activity in vitro were different. Trypanosomes in corresponding ELISA plate wells are reduced and killed. The average mean trypanosome count in the extract was drastically reduced (40.0.0 to 7.0000.33) when 250 g ml-1 of P. biglobosa MPE was used. At 500 g ml-1 of P. biglobosa test extract, trypanosomes were completely killed (40.0.0 to 0.000.00) after 9 hours of incubation, which was statistically equivalent to diminazine aceturate (50 g ml-1) after 4 hours. With a significant difference (P (le) 0.05 to 0. 01), trypanosome counts dropped in a concentration and time-dependent manner. Except at doses of 12.5- 1.56 g ml-1, MPE of P. biglobosa and diminazine aceturate, a typical medication, were cytotoxic to Vero cells. Alkaloids, flavonoids, anthraquinones, tannins, phlobotannins, and cardiac glycosides have already been identified from P. biglobosa leaves and other sections of the plant, and these compounds could be responsible for the plant's increased antitrypanosmal activity.
