Repurposing Siderophores as Antitubercular Agents | Chapter 14 | Emerging Trends in Disease and Health Research Vol. 9

Background: Alternative or supplementary medications are urgently required due to the worrisome issue of drug-resistant Mycobacterium tuberculosis (M.tb). Repurposing already available, FDA-approved medications might help achieve this. In this work, drug-resistant M. tb isolates were used to assess the anti-tuberculosis efficacy of microbial siderophores. On mammalian cell lines, the siderophores' cytotoxicity toward cells was also assessed.

Methods: Mycobacteria Growth Indicator Tube drug susceptibility testing was used to evaluate the anti-tubercular activity of the siderophores. The MTT test was used to assess the siderophores' cytotoxic effects on mammalian cells.

Results: The siderophores tested were shown to be efficient both on their own and when combined with main or secondary anti-TB medications that are often recommended but ineffective against drug-resistant M.tb isolates. Interestingly, When siderophore MIC was evaluated in conjunction with anti-tubercle medications, it fell. There was no discernible cell cytotoxicity seen.

Siderophores are safe supplements to the anti-TB treatment regimen that may be helpful in overcoming drug-resistant tuberculosis.
