A Simple Electrochemical System to Evaluate Interactions between Bauhinia monandra Leaf Lectin (BmoLL) and Cratylia mollis Seed Lectin (Cramoll) Immobilized in Nafion Nanopores with Pseudomonas aeruginosa Endophytic Strain | Chapter 1 | Recent Research Advances in Biology Vol. 9

 The interaction of lectins immobilised in Nafion nanopores with Pseudomonas aeruginosa strains was studied using an electrochemical device. BmoLL, a leaf lectin specific for galactose obtained from Bauhinia monandra leaves, interacted with P. aeruginosa strain UFPEDA598, an endophytic bacterium isolated from B. monandra leaves. Cramoll is a seed lectin from Cratylia mollis that is selective for glucose-binding proteins. In the presence of galactose, a lectin inhibitor, the effect of this interaction was an increase in electrochemical potential and stability, with a binding preference for endophytic membrane carbohydrates. Cramoll had no interaction with the endophytic strain since the electrochemical potential did not differ from the control; nonetheless, UFPEDA-39 (IT2633); after the contact, a reduction in electrical potential was detected, with stability to the glucose-inhibited lectin. In order to recognise microorganisms in real time, we devised a simple, label-free approach. The biosensor was able to detect P. aeruginosa glycoconjugate surfaces from a variety of settings, making it helpful for selective identification.
