A 100% Aqueous Mobile Phase HPLC-photodiode Array Detection of Tetracycline and Its 4-epimer, 4-epi-tetracycline | Chapter 1 | Current Advances in Chemistry and Biochemistry Vol. 9

 This work describes an organic solvent-free mobile phase HPLC method for tetracycline (TC) and 4-epi-tetracycline analysis (4eTC). An InertsilR WP300 C4 column with an isocratic aqueous mobile phase and a photodiode array detector was used to achieve chromatographic separation (PAD). The sample run time was about 5 minutes. Linearity, range, and system appropriateness, the three primary validation performance characteristics, were all well within the international suggested criteria. TC and 4eTC had detection limits of 0.005 and 0.014 g/mL, respectively. The HPLC-PAD system's great repeatability, speed, and ease of use should be useful for assessing TC and 4eTC residues in animal-derived foods.
