Recent Study on Copper-Promoted One-Pot Approach: Synthesis of Benzimidazoles | Chapter 6 | Current Advances in Chemistry and Biochemistry Vol. 3

 For the production of various 2-arylaminobenzimidazoles, a simple, one-pot method was produced. For the first time, a copper catalyst promoted domino C–N cross-coupling reaction for the generation of 2-arylaminobenzimidazoles is used in this process. The synthetic pathway involves a copper-based desulphurization/ nucleophilic substitution followed by domino intra and intermolecular C–N cross-coupling reactions, according to mechanistic studies. This newly developed copper-catalyzed method tackled some of the issues commonly encountered during the synthesis of 2-arylaminobezimidazoles, such as the use of costly catalytic systems and the poor reactivity of bromo precursors. The reaction procedure is straightforward, with good substrate tolerance, and yields of the desired products range from good to excellent. Copper, on the other hand, is more practical due to its simplicity, environmental acceptability, and cost effectiveness.
