Study on Proteomic Profiling of High-grade Glioblastoma: Aspects of Biomarkers | Chapter 8 | Current Advances in Chemistry and Biochemistry Vol. 3

 Using two-dimensional electrophoresis (2DE), mass spectrometry (ESI LC-MS/MS), and immunodetection, researchers were able to identify and quantify different proteoforms (protein species) present in a cell line derived from a high-grade glioblastoma cell line. It was possible to obtain a 2DE protein map with a large data set of 937 spots and 1542 separate protein identifications (proteoforms) from 600 genes. In addition, 16012 proteoforms encoded by 4050 genes were characterised by MS/MS based on their location in 96 gel sections in a separate series of experiments (pixels). Proteins that could be used as glioblastoma biomarkers have received special attention. The list of biomarkers was compiled based on research. The graphs with theoretical and experimental knowledge about proteoforms coded by the same gene were then developed. The condition of these gene products could be better visualised with such a virtual-experimental representation. Many proteins, including possible glioblastoma biomarkers, have a large number of protein species. We believe that these organisms may be a source of highly specific glioblastoma biomarkers.
