A New (Alternative) Model of Heart Lesion in Rat: A Step to Cardiomyoblast Grafting for Cardiac Tissue Repair | Chapter 3 | Highlights on Medicine and Medical Research Vol. 4

 An experiment with 57 rats was carried out in order to create a new model of cardiac lesion.

With a «Cautery high temperature fine tip» (Bovie Medical Corporation-USA), the antero-apical region of the heart (i.e. a portion of the right, septum, and left ventricles) was cauterised to establish an injury of controllable, precise, and repeatable dimensions (8x8 mm2 surface, 1 mm depth).

Electrocardiography, pulse oximetry, echocardiography, and optic microscopy were performed at various times (from day 0 to day 230) after the procedure to determine the functional and morphological characteristics of the lesion.

After some technological tweaking, the last 12 run animals had a 100% survival rate. ST-modifications on the ECG in two leads, a substantial decrease in the ejection fraction (37.4 percent at day 10+/-2 and 27.3 percent at day 30+/-5), necrotic changes, and fibrosis of the lesion region in the myocardium) were all recorded.

The fact that the injury survived and progressed indicates that the proposed model may be useful for research into cardiac tissue repair.
