Use of Rapd and ISSR Polymorphisms for Detection of Molecular Diversity of Date Palm (Phoenix dactylifera L.) Cultivars from Algeria | Chapter 9 | Advanced Research in Biological Science Vol. 3

 This division investigated the ISSR and RAPD polymorphisms in date touch cultivars collected from Bou-Saada oasis, situated at the bases of the Ouled Nail Range of the Saharan Atlas of Algeria. Date palm (Phoenix dactylifera) is an main crop plant both from nutritional and financial points of view. The assessment of hereditary diversity and people differentiation of date palms are evaluative for its vital conservation and tenable utilization of its ancestral diversity. Samples were composed from young leaves of adult trees and retained frozen with liquid nitrogen. DNA ancestry was performed in accordance with the standard protocol. A total of 198 (RAPD) and 151 (ISSR) fragments were amplified and nick via electrophoretic gels. Fig. 2 shows instances of resulted banding patterns in the 20 cultivars for the RAPD textbook OP-N01 and ISSR primer A38. Both markers happened in high and corresponding levels of polymorphisms; around 75% polymorphic positions were detected with 151 ISSR and 198 RAPD alleles. The contribution of RAPD and ISSR indicators to total gene variety was 0.225 and 0.220 and to Shannon's Information index was 0.344 and 0.337, respectively. At the group level, comfortable, dry, and median cultivars are distinguished apiece presence of particular alleles unshared to each group, that is seen in significant alternatives in gene difference as well as Shannon's Information indexes. But rather than clarifying variation 'tween groups, the proportion of prevailing alleles within groups led to forceful variation (100% for ISSR and 98% for RAPD) made clear within groups of individual cultivars. Generally, RAPD and ISSR markers happened in disparate grouping patterns, however, two together markers configured the close relationship 'tween Rotabaya and Ghars cultivars with 84% start operating system value (BV; RAPD) and 63% BV (ISSR). The cultivar, Baarit- djemal is strongly recognized as possible parent of the sampled male tree. Baarit-djemal cultivar and the male forest found closely accompanying with BV equaled to 90% for variety of combined markers, 71% accompanying RAPD markers and 50% accompanying ISSR markers. The assembling patterns resulted from combined dossier, on the other hand, have habitual the relationship among Mekarkcha and Deglet nour accompanying 94% BV compared to that shown with ISSR stones (98%). Cataloguing the existing date palms at molecular level maybe a mandate to support various phases of selection programs and preservation strategies.
