AKSS16-LIV01: An Antioxidant and Immunity-Boosting Multiherbal Formulation for Alcoholic Liver Dysfunction in Experimental Animal Model | Chapter 2 | Recent Developments in Chemistry and Biochemistry Research Vol. 5

High rates of alcohol intake are frequently the cause of liver complications. The majority of alcohol-drinking citizens in industrialized and developing nations suffer from cirrhosis, fatty liver, liver fibrosis, and even hepatocellular cancer, among other liver issues. Creating symptomatic, safe medication to go through this is a new global problem. The primary goal of the research is to provide a symptomatic, safe medicine to lessen liver damage caused by ethanol. We created a multi-herbal formulation (AKSS-16-LIV01), which may be helpful in liver complications.  In this investigation, we try to reduce liver damage from a variety of toxicants. Swiss albino mice were split up into seven groups, and each group was subjected to ethanol-induced injury for several weeks before our herbal preparation was used to investigate a healing response. The groups include the normal control group (Group-I), the group treated with 50% v/v ethanol (Group-II), the groups pre-treated with AKSS16-LIV01 at a low dose of 100 mg/kg/day (Group-III), the group pre-treated with AKSS16-LIV01 at a middle dose of 200 mg/kg/day (Group-IV), the group pre-treated with AKSS16-LIV01 at a high dose of 400 mg/kg/day (Group-V), the group pre-treated with Sylimarin at 100 mg/kg/day (Group-VI), and the only group treated with AKSS16-LIV01 (400 mg/kg/day) (Group-VII).In the mice given ethanol, the results showed a marked increase in a number of biochemical parameters, lipid profile parameters, lipid peroxidation, nitric oxide (NO) concentration, nitric oxide synthase level, and pro-inflammatory cytokines, such as tumour necrosis factor (TNF-\(\alpha\)) and transforming growth factor (TGF-\(\beta\)1). However, ethanol significantly decreased the levels of tissue antioxidant enzyme activity (SOD, CAT, GSH, and GPx), serum total protein, total albumin, albumin globulin ratio, and tissue total antioxidant enzyme activity. The formulation (AKSS16-LIV01) was applied therapeutically at a dose that determined its effectiveness in suppressing all relevant parameters mentioned above, while also shielding the liver from ethanol-induced fibrogenesis. In addition to this gross morphology of the liver, the hepato-protective impact of the formulation in comparison to the usual medication Sylimarin was clearly supported by H&E liver histology and Masson trichrome and serius red analysis of the liver sections. According to the study's findings, a multi-herbal formulation that was designed (AKSS16-LIV01) and given at a dose of 400 mg/kg/day produced the best possible response to lessen ethanol intoxication. The results unmistakably show that AKSS16-LIV01 may be a safe and non-toxic drug that shields the liver from oxidative damage caused by ethanol and keeps levels of pro-inflammatory cytokines stable over time.

Pattern of Dyslipidemia among Prediabetics of Karnataka State, South India: A Cross Sectional Study |Chapter 4 | Medicine and Medical Research: New Perspectives Vol. 6

This study aims to explore the possibility of the existence of dyslipidemia among prediabetics in Karnataka state south India and to identify the risk factor(s) if any, for the onset of diabetes. Hyperglycemia can be determined by the glycated hemoglobin (HbA1c) value which indicates the blood glucose level over the past three months. A cross-sectional study involving 310 individuals in Bangalore, Karnataka State, South India from the period of February 2022 to March 2024 was considered for the study. Within the lipid profile, Total cholesterol (TC), High-Density Lipoprotein (HDL) and the ratio of TC to HDL displayed statistically significant differences between the means of the population of healthy nondiabetics and prediabetics. So, testing of TC, HDL and TC to HDL ratio at regular intervals for prediabetics should be made mandatory, as they fall in the risk category for developing type 2 Diabetes and this regular screening of TC,  HDL and TC to HDL ratio will prevent prediabetics from progressing into type2 diabetes later on. It is a well-known fact that the HbA1c level as a predicting factor differs among races and ethnicities as the rate of glycation and RBC life span differs among racial and ethnic groups. Regarding demographic factors, abdominal obesity was found to be statistically significantly associated with prediabetes. Prediabetics should implement a regular exercise regime to prevent themselves from becoming diabetics in their later years.

Effects of the Vita Slim and Green Tea as Anti-obesity Drugs on Certain Biochemical and Physiological Indices of Obese Adult Male Albino Rats | Chapter 9 | Recent Developments in Chemistry and Biochemistry Research Vol. 8

Background: A larger number and larger size of fat cells characterize obesity, a condition that has a detrimental effect on one's health. Because obesity is linked to an increased risk of diabetes and heart disease, the ideal anti-obesity medication would have weight loss that was long-lasting and had few side effects.

Objective: The objective of the study is to detect if the weight-loss drugs; green tea and Vita Slim negatively affect several biochemical and physiological markers.

Materials and Methods: This study utilized a total of twenty-one albino male rats (220±20 g). Rats were purchased from an animal unit of the Nile Pharmaceutical Company, Cairo, Egypt. Three groups consisted of 21 mature obese male albino rats (with a body weight of 220 ±20 gram): In Group one: control (vehicle treatment), in group two: for a period of 30 days, rats were given Vita slim (0.1mg/kg/day), and in group three: for 30 days, rats were given green tea (0.1 mg/kg/day).

Results: Significant changes were observed in the weight of the treated groups, in serum levels of AST/ALT, albumin, triglycerides, low-density lipoproteins (LDL), LDL/HDL ratio, urea, creatinine, as well as triiodothyronine (T3) as compared with the control group. It is proven that thyroid hormones, particularly T3, are important metabolic hormones. Energy generation is linked to the quantity of thyroid hormones in the bloodstream.

Conclusion: The study concluded that Vita Slim and green tea capsules have a deleterious influence on critical biochemical and physiological markers, notably liver functions. Several physiological and biochemical markers, including kidney and liver functions, have been shown to be negatively impacted by Vita Slim and green tea after 30 days of ingestion.

Evaluating the Impact of Glycemic Control, Lipid Derangement and Antioxidant Status on Progression of Diabetic Retinopathy | Chapter 11 | Achievements and Challenges of Medicine and Medical Science Vol. 8

Background: Diabetes Mellitus is a global health problem. It is a potentially life-threatening metabolic syndrome. There were nearly 171 million diabetics worldwide in 2000 with the number projected to be almost 2.5 times, about 366 million by the year 2030 (prevalence rate of 4.4%) with the greatest number of cases expected in China and India. Patients with diabetes show a difference in serum uric acid pattern in relation to the duration of the disease along with glycemic status, lipid derangements and complications associated with the disease.

Aims and Objectives: To study HbA1c levels to know the blood sugar levels for the past 3 months to assess the glycemic control and to estimate lipid profile to assess the risk of microvascular complications like Retinopathy in diabetic cases. Serum uric acid levels are estimated to evaluate its role in the pathophysiology of diabetes and to establish its association with glycemic control and lipid profile which helps to understand the onset and progression of complications such as retinopathy.

Materials and Methods: The present study consists of 75 cases of chronic diabetes with retinopathy as a complication in the age group of 45-75 years. The values of HbA1c, lipid profile and uric acid in cases are compared with that of 50 apparently healthy non-diabetics who are of the same age group. All the subjects were from the ophthalmology department of Govt. General Hospital, Kakinada and also from Nayana Eye Care Hospital, Kakinada.

Results: The Mean Values of HbA1C and lipid profile were significantly high in diabetic retinopathy cases (p<0.0001) as compared to the control group. The Mean values of uric acid in diabetic retinopathy cases do not show a significant difference from that of the control group (p=0.446).

Conclusion: The study shows that poor glycemic control plays a major role in the onset and progression of diabetic retinopathy. The significant changes in the lipid profile levels in diabetic retinopathy indicate lipid peroxidation plays a key role in the progression of the disease. Serum uric acid level has not shown statistically significant changes in diabetic retinopathy implying no role of uric acid in the progression of disease.

Association between Serum Ferritin and Some Bio Variables in Obese Male and Female at Obesity Medical Center in Iraq | Chapter 3 | Disease and Health: Research Developments Vol. 5

Obesity is a worldwide health complication. In 2014, 600 million adults and 42 million kids, less than five years of age, were obese. It is a hyperassembly of energy in the form of adipose tissues which has worse effects on health. The major factors of the obesity epidemic are: heavy meal which is rich in lipids, carbohydrates resulting in overweight and eventually obesity, and a decrease in physical activity that leads to defects in calorie conception. In health care facilities ferritin assay is used as a screening test to detect iron deficiency; however, its reliability in obesity has been questioned. The present study aimed to assess and compare the incidence of obesity, hyperferritinemia, hyperlipidemia, and hypouricemia among middle-aged hypertensive patients in the local setting and examined ferritin concentrations and other classical indices in obese males and females. Serum levels of ferritin was assessed using ELISA kit, while Hb was analyzed using Coulter LH 750 Hematology Analyzer and other parameters were assessed by colorimetric kits. This limited population study was conducted at the Obesity Medical Center at Al-Kindy Teaching Hospital, Baghdad, Iraq from January to March 2016. One hindered and eight patients were enrolled in this study plus to participate of 61 subjects as healthy control. Based on body mass index (BMI), 169 participants were divided into two groups: BMI ≥ 18.5–25 kg/m2 non obese (control), BMI ≥ 30 kg/m2 obese subjects. Patients with endocrine disease, pregnancy, and renal disease leading to hypertension were excluded from the study. The statistical package for the social sciences computer program version 20 (IBM SPSS Statistic software, IBM Corporation, New York, United States) was used for the analysis. The main demographic features data of the study population shows that systolic blood pressure and diastolic blood pressure were raised significantly in obese patients (P<0.05) compared with control. Serum hemoglobin levels were decreased significantly in obese males when compared with the non-obese group and no difference in the female group was observed. In addition, the levels of lipid profile and other studied parameters according to gender showed the following results:- significant elevation in TG, LDL, VLDL, glucose, and ferritin in the obese (for both genders) compared with the non-obese group. A significant decrease was noticed in HDL-c, uric acid, and urea levels in the obsessed group compared with the non-obese group in both genders. The comparison results between obese males and females concluded that a significant elevation in BMI with a significant decrease in VLDL and ferritin levels in obese females compared with obese males. The present study examines all types of correlations between parameters. In obese patients (n=108), stepwise linear regression analysis showed that serum ferritin levels were independently correlated with LDL, VLDL levels, and age, (P< 0.001). Increased BMI and ferritin levels are more frequent in females as compared to male obese patients which are an important risk factor for obesity complications.

Effect of Habitual Consumption of Madhuca longifolia Drinks on Diabetes and Dyslipidemia among Santhal Tribals | Chapter 3 | Advanced Concepts in Pharmaceutical Research Vol. 4

This affiliate aims to investigate the Effect of usual consumption of Madhuca longifolia drinks on diabetes and dyslipidemia with santhal tribals. Madhuca longifolia (mahua) is a tropical tree extensive throughout India, generally on plains.  By way of its abundant therapeutic benefits, the tree is visualized as a godsend by tribals the one live in the woodland.  Further studies proved that advantageous effects are observed in Santhals many of whom are balanced consumers of Madhuca longifolia drinks. The authors still did a study on this facet.Diabetes and lipid profiles of ancestry, as well as normal anthropometric parameters, were assessed in a Santhal family population (only men were recruited), many of whom, but not all, are established mahua drink consumers. The results were statistically resolved and compared using the undergraduate's t-test, and conclusions were attained. The results demonstrate that anthropometric measures do not vary much, but FBS does. HbA1c, cholesterol, and LDL cholesterol levels are reduced with mahua consumers compared to non-shoppers. When compared to the additional group, mahua consumers have bigger HDL levels. In the case of VLDL and triglycerides, no significant effect was discovered. The research found that common mahua drinks among the Santhal tribe enhanced diabetes and lipid profiles, other than triglycerides and VLDL.  More extensive research is necessary to get correct insight and practical benefits concerning this.

Determination of Lipid Profile and Atherogenic Indices for Cardiovascular Disease Risk Based on Different Fish Consumption Habits| Chapter 7 | Challenges and Advances in Pharmaceutical Research Vol.1

The ingestion of moderate amounts of fish on a regular basis is linked to a lower risk of death from cardiovascular disease (CVD). However, due to the oxidation susceptibility of the polyunsaturated fatty acids in fish, the positive effects of a fish-enriched diet appear to be paradoxical. The atherogenic index is a useful measure for identifying patients who are at risk for cardiovascular disease. Certain fish dietary habits have a positive effect on lipid profile levels, lowering the risk of coronary heart disease. The goal of this study was to assess CVD risk using atherogenic indices and to examine the plasma lipid profile of healthy persons with various fish intake habits.

Methods: Healthy people's fasting blood samples were taken for lipid profile calculations using an automated analyzer. Each participant provided detailed information on their physical and atherogenic characteristics.

Conclusions: The CVD risk profiles varied depending on the type of fish consumed. Total cholesterol, triglyceride (TG), and non-high-density lipoprotein (Non-HDL-C) levels differed substantially (p0.05), whereas high-density lipoprotein cholesterol levels differed considerably (p0.001). Inland fish consumers had a higher level of TG/HDL. The atherogenic coefficient and the Castelli risk index-2, on the other hand, did not differ significantly depending on the type of fish consumed. All fish eaters, including beef eaters, had atherogenic indices that were borderline "high risk."

Conclusion: People who ate fish and beef were at a higher risk than those who ate inland and sea fish. Non-HDL cholesterol may be a marker for a lipid profile in the blood linked to an elevated risk of heart disease. Non-HDL-C levels were borderline high risk for CVD in all fish, including beef eaters. There were age and sex-related differences in the TC and lipid-protein fractions. TC and LDL-C values were greater in women than in men. Non-HDL-C and TG/HDL-C levels can be utilised as more accurate indicators of CVD risk.

Investigation on the Anti-ulcerogenic Effects and Anti-oxidative Properties of Ceiba pentandra Leaves on Alloxan-induced Diabetic Rats | Chapter 8 | Trends in Pharmaceutical Research and Development Vol. 5

It has been shown that many natural products, mainly of plant origin, have promising practises that could help prevent and/or improve disease prevention. Gastric ulcer is a gastrointestinal tract ulceration arising from repeated erosions and stomach wall damage that can become perforated and develop into peritonitis and massive haemorrhage. Objectives: The current research aimed to assess the protective effects of Ceiba pentandra leaves methanol extract on gastric ulcers induced by indomethacin and ethanol and on oxidative stress indices of diabetic rats induced by alloxan. Location and length of study: Department of Biochemistry, University of Nigeria, Nsukka. Nature of the study: extraction and administration of graded doses of the extract. State of Enugu, Nigeria, from May, 2011 through October, 2011. Methodology: Ceiba pentandra leaves were extracted using methanol. For each of the ulcer experiments, 20 adult rats, split into five groups of four rats each, were used. In rats, gastric ulceration was caused by oral administration of indomethacin (50 mg / kg) and 95% ethanol (0.5 ml) thirty minutes after extract therapy and sacrificed to animals 8 hours later. Thirty (30) albino rats, divided into six (6) groups of five (5) rats each, were used for the diabetes research. Diabetes was induced by i.p. injection of alloxan monohydrate (150 mg / kg) for two weeks in overnight fasting animals and animals treated with a range of extract doses (100 , 200 and 400 mg / kg). For the determination of lipid profile and liver marker enzymes, serum obtained from the diabetic rats was used. Results: inhibition of severe and dose-dependent ulcers (70, 82 and 84 percent; 19, 53, and 58 percent for 100 , 200, and 400 mg / kg extracts, respectively) were developed for the ulcer models used in all extract-treated classes. Important reductions (p<0.05) were reported in fasting blood glucose, liver marker enzymes, total cholesterol, low density lipoprotein and triacylglycerides relative to those of the diabetes-untreated community in the serum of the extract-treated groups. Conclusion: The results of this study indicate that Ceiba pentandra leaves methanol extract has good anti-ulcerogenic and anti-oxidative properties and has the potential to be used as a herbal remedy for the treatment of gastrointestinal ulcers and diabetes control.
