Two distinct reversed-phase high performance chromatographic methods were developed for the simultaneous determination of naringin, hesperidin, and organic acids in natural and commercial citrus juices. For naringin and hesperidin, the separation was achieved on a C18 column using an acetonitrile: water: formic acid (21:78.8:0.2) solution at pH 2.5 as a mobile phase under isocratic conditions. The analysis was performed at room temperature with a flow rate of 1 mL.min-1 and a total run time of 15 minutes. The wavelength of detection was set to 280 nm. Organic acids were separated chromatographically in a single run using isocratic mobile phase containing 10 mM. KH2PO4 (pH=2.2) at room temperature with a flow rate of 1 mL.min-1 on a C18 column in 10 minutes. The ultraviolet absorption was measured at 210 and 245 nm. Calibration linearity, accuracy, and precision were all investigated as part of the method validation. The improved method was used to analyze natural and commercial citrus juices. When compared to natural citrus juices, commercial fruit juices had lower levels of naringin, hesperidin, and organic acids.
