Evaluating the Antioxidant Potential and High Performance Thin Layer Chromatography Profiling of Siddha Formulation Veppampoo Mathirai | Chapter 7 | Medical Science: Trends and Innovations Vol. 7

In today’s world, herbals play a significant role in boosting immunity and preventing diseases. In some parts of the world, they are the primary required materials for healthcare system. In herbal formulation, some plants or specific combinations of herbs may act as antioxidants by exerting superoxide scavenging activity or by increasing superoxide dismutase activity in various tissue sites. Plant antioxidants play a vital role in the battle against cellular damage and disease. The present study aimed to study the antioxidant potential and basic TLC, HPTLC fingerprinting profile of poly herbal Siddha formulation Veppampoo Mathirai used for hypertension to regulate blood pressure in the Siddha system of medicine. The antioxidant action was measured by ABTS free radical scavenging assay, lipid peroxidation and nitric oxide scavenging measure. The common modern techniques that help to evaluate the quality control of complex herbal medicine are thin layer chromatography (TLC) and High performance thin layer chromatography (HPTLC). World Health Organization also indicated the above as a valuable tool to ensure the quality of medicinal plant products. Analysis of TLC was done by silica gel 60F254, 7X6 cm (Merck) and HPTLC by CAMAG Twin trough chambers. All outcomes of the sample were communicated as mean ± standard deviation compared with control. The study revealed the presence of a prominent peak at 366 nm in TLC analysis and twelve prominent peaks in HPTLC analysis with Retention factor (Rf) value ranging from 0.02 to 0.86. ABTS scavenging assay, lipid peroxidation and nitric oxide scavenging showed EC50 values of aqueous extract of Veppampoo mathirai at 55.32 μg/mL, 51.3 μg/mL and 51.31 μg/mL, separately. The above results revealed that the aqueous extract of Veppampoo mathirai has solid antioxidant possibilities and is also a good source of natural antioxidants. It is concluded that Siddha formulation Veppampoo mathirai is a good source of natural antioxidants that can be utilized for age-related diseases like blood pressure as therapeutic.

Chromatographic Finger Printing Analysis in Identification of Phytomarkers by HPTLC Technique | Chapter 9 | Advanced Concepts in Pharmaceutical Research Vol. 2

 Chromatographic fingerprinting methods are becoming more popular and significant in the uniformity of natural fruit. They are employed for two together quantifying marker compounds and authenticating the presence of differing herbs in poly herbal arrangements. Although HPLC and GC are also frequently working for chromatographic fingerprinting of single spice and poly herbal arrangements, HPTLC has been the pioneer in this field. Chromatographic fingerprinting is used to decide the single herbs labeling and qualities before they are contained to herbal formulations. Each herb fated in near future incorporated to the expression must first undergo fingerprinting. This maybe used to identify both the spice and evaluate the main alive components which maybe found in the spice. The final poly herbaceous formulation is developed when all the herbs have happened appropriately treated and their identity and innocence have been confirmed. At this point, a chromatographic means can yield distinctive peaks each of the herbs utilized in herbal expression is used to develop a chromatographic mark of the poly herbal expression. Estimation of Metformin Hydrochloride, Gliclazide and Pioglitazone Hydrochloride: A UDDD-HPTLC Densitometry Method Validation Study | Chapter 12 | Novel Aspects on Pharmaceutical Research Vol. 3

Investigation of Hypolipidemic Activity of Nanoparticles Containing Ethanolic Extract of Cocculus pendulus in High Fat Fed Rats | Chapter 2 | Challenges and Advances in Pharmaceutical Research Vol. 2

Cardiovascular disease is caused by an abnormal presence of lipids in the circulation (CVD). The major goal of this work was to make nanoparticles from an ethanolic extract of Cocculus pendulus and test their hypolipidemic efficacy in rats fed a high-fat diet. Preliminary phytochemical examination of the complete plant of Cocculus pendulus indicated the presence of alkaloids, phytosterols, flavonoids, triterpenes, and volatile oils, among other phytoconstituents. HPTLC fingerprinting of an ethanolic extract of the plant Cocculus pendulus displays 13 peaks at 256 nm and 10 peaks at 366 nm, respectively, while GC-MS analysis demonstrates the presence of -Sitosterol as a marker component. The use of nanotechnology to improve the bioavailability of herbal medicines is a promising strategy. The optimal nanoformulation revealed a mean particle size, polydispersity index, and zeta potential of 536 nm, 0.256, and 16.4 mV, respectively, of ethanolic extract of Cocculus pendulus nanoparticles prepared using the Solvent evaporation approach. On male Wistar rats, antiatherogenic efficacy was tested using ethanolic extract dosages of 200, 400 mg/kg and optimised nanoparticles doses of 40, 80 mg/kg. The effects of two different dosages of ethanolic extract of Cocculus pendulus and nano formulations on an 8-week high-fat diet were investigated. Finally, results show that when Cocculus pendulus and Nano formulations were administered orally at two dose levels, there was a significant reduction in body weight, total cholesterol, low density lipoprotein, phospholipids, triglycerides, and VLDL levels, as well as a similarly significant increase in high density lipoprotein levels. Cocculus pendulus nanoparticles with doses of 40 mg/kg and 80 mg/kg had better hypolipidemic action than the ethanolic extract of Cocculus pendulus with doses of 200 mg/kg and 400 mg/kg.

Studies on Anti-Inflammatory and General Glucocorticoid Physiology in Skeletal Muscles Affected by Duchenne Muscular Dystrophy: An approach towards Exploration of Steroid-Sparing Agents | Chapter 4 | Current Advances in Chemistry and Biochemistry Vol. 7

 The activation of proinflammatory and metabolic biological pathways in skeletal muscle cells is a hallmark of Duchenne muscular dystrophy (DMD). The dystrophin-associated protein complex, a large transmembrane protein complex that acts as a shock absorber during muscle contraction, a receiver and transducer of cellular signals, and a scaffold for signalling proteins, is destabilised in the absence of dystrophin, a protein that provides a link between the extracellular matrix and the myocyte cytoskeleton. The majority of these pathways are inhibited by synthetic glucocorticoids. Hypertension, arrhythmias, hyperglycemia, osteoporosis, weight gain, growth retardation, skin thinning, cushingoid look, and tissue-specific glucocorticoid resistance are all side effects of glucocorticoids. As a result, reducing the glucocorticoid dosage for DMD patients may be advantageous. When looking for compounds that can accomplish similar pathway stabilisation in DMD, a deeper understanding of the primary cellular pathways that are stabilised following synthetic glucocorticoid administration is required. This review gives a quick rundown of the primary anti-inflammatory pathways as well as the metabolic effects of glucocorticoids in DMD-affected skeletal muscle. The known medicines that can stabilise these pathways and could be used as steroid-sparing medicines in combination with glucocorticoid therapy are outlined. This could open up new avenues for research in DMD animal models and/or human trials, potentially resulting in lower glucocorticoid dose regimens for DMD patients.

Determination of Telmisartan and Hydrochlorothiazide Using HPTLC in Human Plasma: Development and Validation of Bioanalytical Method | Chapter 3 | Current Advances in Chemistry and Biochemistry Vol. 7

 A simple, sensitive, quick, and cost-effective thin layer chromatographic technique was used to determine the concentrations of telmisartan and hydrochlorothiazide in human plasma using paracetamol as an internal reference. A combination of hydrochlorothiazide (HCT) and telmisartan (TEL) is more successful than either medicine alone in the treatment of hypertension.

The plasma sample was collected using a methanol-acetonitrile (3.0:0.1, v/v) mixture. Concentrations of 200, 400, 600, 800, 1000, and 1200 ng/spot were employed for the hydrochlorothiazide and telmisartan calibration curves, respectively. Telmisartan and hydrochlorothiazide, respectively, showed 75.98 percent and 81.91 percent recovery. Chloroform: methanol: toluene (8:2:4 v/v/v) makes up the mobile phase. At a wavelength of 278 nm, densitometric analysis was performed. The Rf values for hydrochlorothiazide, paracetamol, and telmisartan, respectively, were 0.28 0.05, 0.50 0.05, and 0.66 0.05. The stability of telmisartan and hydrochlorothiazide in plasma was validated in three freeze-thaw cycles (20 C), on bench for 24 hours, and post preparative for 48 hours. A recovery study was used to statistically validate the recommended method for identifying telmisartan and hydrochlorothiazide in human plasma. The method is less expensive and faster than previously disclosed approaches. In the future, we may be able to use this technique for bioequivalence research.

Assessment and Quantification of Pharmacologically Active Marker Gallic Acid and Ellagic Acid from Leaf and Stem of Pergularia daemia Forsk. by HPTLC Method | Chapter 13 | Technological Innovation in Pharmaceutical Research Vol. 1

 A TLC densitometry method is developed for the quantification of gallic acid and ellagic acid using HPTLC. HPTLC is an important tool for determining chemical constituents in plant material. This is the first time that these two biologically active compounds, gallic acid and ellagic acid, have been quantified using HPTLC from this plant. Gallic acid and ellagic acid were quantified from the methanol extract using the Toluene: Ethyl acetate: Formic acid (6:4:0.8 v/v/v) solvent method. Gallic acid and ellagic acid have Rf values of 0.43 and 0.38, respectively. Gallic acid (100 to 600 ng) and ellagic acid (100 to 600 ng) have identical linearity levels, with correlation coefficients (r- values) of 0.9994 and 0.9997, respectively. Gallic acid and ellagic acid concentrations were 3.01 and 11.09 g/ml in the leaf and 4.97 and 18.30 g/ml in the stem, respectively. Gallic acid and ellagic acid quantification demonstrated strong separation and resolution from other extract constituents. The flexibility, precision, and selectivity of this approach are its key advantages. Its key benefits are its ease of use, precision, and selectivity. This approach can also be used to estimate these compounds in other herbal preparations, and it can help with standardisation.
