The grouper Epinephelus lanceolatus is a vulnerable species of high economic value, so this study aims to determine the effect of the combination of 6% glycerol with different egg yolk concentrations on the post-cryopreservation quality of the grouper Epinephelus lanceolatus spermatozoa (Bloch, 1970). Using the stripping process, the grouper giant semen fish was collected. The diluent is made of Marine Fish Ringer, 6 percent glycerol and various egg yolk concentrations (0 percent ; 5 percent ; 10 percent ; 15 percent ; 20 percent ; and 25 percent ). The equilibration was carried out for 10 minutes at 4oC, and freezing was carried out for 48 hours at (-20oC) using a freezer, as well as defrosting for 30-60 seconds at 45oC. The semen was measured macroscopically (color, volume and pH) and microscopically (motility, viability and abnormality) as well as the ability to fertilize the grouper eggs of the tigers. There was a substantial difference (p< 0.05) in motility, viability and ability to fertilize post-cryopreserved grouper spermatozoa based on the ANOVA and Tukey test, but there was no significant impact on maintenance after post-cryopreserved abnormalities of the spermatozoa (p> 0.05). The findings showed that 15 percent of the treatment of egg yolk was the optimum concentration since 83.64 ± 1.72 percent, 81.44 ± 2.06 percent and 77.31 ± 1.90 percent, respectively, provided the highest mean percentage values of motility, viability and fertilization capacity. The number of anomalies of 21.50 ± 1.20 percent is the lowest average value.
