Rutin Attenuates Symptoms Associated with Endotoxemia by Regulating LPS-Induced Pro-Inflammatory Cytokines | Chapter 2 | Novel Aspects on Pharmaceutical Research Vol. 9

 Background: Rutin, a open flavonoid compound widely distributed in buckwheat, salads and fruits, is a well-known for a type of biological actions and pharmacological effects.Objectif: The present study proposed to explore whether Rutin keep attenuate lipopolysaccharide (LPS)-persuaded inflammatory answers in an experimental endotoxemia. Methodology: We investigated the attainable effects of Rutin against LPS-persuaded endotoxemia model in vivo to determine either Rutin could rescue mice from resultant death afterwards their exposure to LPS. Forty-eight mice were detached into four groups (n=12/group) and challenged accompanying LPS (5–40 mg/kg) via a alone intraperitoneal (IP) injection. The mice were noticed for mortality doubly a day for 7 days, and continuation rates were recorded. Mice were treated accompanying RT at different doses (60, 80 or 100 mg/kg), or bicycle (5% of DMSO in saline), for one intraperitoneal (i.p.) route. One hour later, animals were disputed with LPS (20 mg/kg). The reactions of cytokines were also evaluated in serum isolated from ancestry collected at 0, 2, 4, 6, 8, and 24 h subsequently LPS administration of the rodent.Results: The study indicates that Rutin showed suppressing effects on TNF- α, IL-6, and IL-1 β result by LPS- challenged rodent. Furthermore, RT protected mice by reconstructing host survival against LPS challenge.Conclusion: The verdicts demonstrate that rutin offers guardianship against LPS-induced endotoxemia and will be potentially valuable as an adjuvant situation for inflammatory diseases.

Optimization Extraction Conditions of Some Flavonoids from Elsholtzia ciliata Leaves by Response Surface Method | Chapter 4 | Current Overview on Pharmaceutical Science Vol. 9

 In this study, the distillation conditions of various flavonoid compounds from Elsholtzia ciliata leaves were optimized for one response surface means (RSM). The experiment was arranged in accordance with the Box - Behnken design accompanying 15 experiments and was built established an optimized process for the ancestry of ethyl gallate (Y1), rutin (Y2), apigenin (Y3), and quercetin (Y4) with 3 determinants of extraction hotness (X1), extraction period (X2) and pressure (X3). The conditions for optimizing the ancestry process to obtain the best content of some flavonoid compounds were at 57O C, 54 notes of meeting, and 100 bar pressure. The experimental principles at the optimal limits of ethyl gallate, rutin, apigenin, and quercetin are 97.65 ± 0.32 mg/100g, 35.50 ± 0.25 mg/100g, 17.62 ± 0.15 mg/100g, and 27.66 ± 0.20 mg/100g, respectively.

Chromatographic Analysis Technique for the Determination of Phenolic Compounds in Brassica oleracea L.var capitata | Chapter 6 | Challenges and Advances in Pharmaceutical Research Vol. 8

 Brassica foods are particularly nutrient-dense, offering vitamins, carotenoids, fibre, soluble sugars, minerals, glucosinolates, and phenolic compounds, among other nutrients and health-promoting phytochemicals. The current knowledge indicates that the aging processes may result from oxidative stress leading to a variety of alterations within the human organism caused by Reactive Oxygen Species (ROS). In the present work, high-performance liquid chromatography was used to assess the phenolic compounds rutin, quercetin, and kaempferol in Brassica oleracea L.var capitata (HPLC).For the estimation of Rutin, Quercetin and Kaempferol by HPLC method, the mobile phase used are, Acetonitrile and Phosphate buffer (pH=5.8) in ratio of 55: 45. Quantification of Rutin, Quercetin and Kaempferol was carried by Athena C18 column and absorbance was measured at 254 nm with flow rate of 1 ml/min. In HPLC analysis the retention time (Rt) of standards, Rutin, Quercetin and Kaempferol were found to be 2.357, 6.093 and 9.373 respectively, while the Retention times of Rutin and Kaempferol in Brassica oleracea L.var capitata are 2.387, 6.060 and 9.113 which are found to be matching with standards retention time values respectively. In comparison to other approaches, the cost and running time per analysis are determined to be cheap. So, rutin, quercetin, and kaempferol can all be quantitatively analysed using this method. The procedure was discovered to be quick, effective, and straightforward and can be used as quality control for phenolic chemicals             (rutin, quercetin, and kaempferol).

Traditional Medicine Heteropterys cotinifolia: A Neuropharmacological and Phytochemical Approach with Possible Taxonomic Implications | Chapter 4 | Emerging Issues in Science and Technology Vol. 4

Heteropterys cotinifolia (Malpighiaceae) has been used in traditional Mexican medicine mainly for the treatment of nervous disorders. However, the specific neuropharmacological activities responsible for this use remain to be defined. The present study evaluates the antidepressant and anxiolytic effects produced by the methanolic extract of Heteropterys cotinifolia and the influence of such effects on motor activity in ICR mice. Our results show that the methanolic extract of Heteropterys cotinifolia produces a dose-dependent antidepressant effect in the forced swimming test in mice at doses from 31 to 310 mg/kg, with no reduction of mice locomotion. However, no anxiolytic properties were observed.  Our findings suggest that the main extract compounds identified as chlorogenic acid and rutin may be involved in the antidepressant effects. The presence of flavonoids in the methanolic extract of Heteropterys cotinifolia may also provide further data to characterize taxonomically this species in order to be distinguished from others species closely related and belonging to the same genus.  According with recent reports, antioxidant and anti-inflammatory properties could be involved in the progression and chronicity of various CNS diseases and cancers. Since the fact that other species from the genus Heteropterys possess outstanding antioxidant properties and the flavonoid content finding in this extract, we continue our investigations on the methanolic extract of   Heteropterys cotinifolia from which, the flavonoid 5,4´-dihydroxy-3,6-dimetoxiflavone was isolated. The extract and flavonoid showed significant antioxidant effect  in DPPH and TBARS assays, while IC50 values for extract (24.46±5.96) (25.92±0.56) and flavonoid (9.54±0.14) (35.88±2.24), respectively, were higher than references. Flavonoid exhibited a high percentage of inhibition (>50%) against the total cancer cell lines evaluated, contrasting with extract (<11.90%). However, no antiinflammatory activities were observed in TPA-induced ear edema in ICR-strain mice. Taking all this information together, Heteropterys cotinifolia possesses additional to neuropharmacological activities
