Febuxostat (FBS) has emerged as a revolutionary treatment for hyperuricemia and gout. This comprehensive review provides a pioneering analysis of the instrumental role of Liquid Chromatography with Mass Spectrometry (LC-MS) in unravelling the bioavailability and bioequivalence of Febuxostat (FBS) in human plasma. Utilizing Febuxostat d9 and d7, along with other drugs as internal standards (IS), the review steers through the intricate view of pharmaceutical research, focusing on validation methods and the challenges presented by the complex pharmacokinetics of FBS. Unprecedented in its depth, the review aims to contribute valuable insights to the field by addressing the evolving paradigms in pharmaceutical analysis. FBS, a revolutionary advancement in the treatment of hyperuricemia and gout, takes centre stage, highlighting its pivotal role in contemporary therapeutic approaches. LC-MS develops as a cornerstone analytical method for studying FBS, offering unparalleled sensitivity and selectivity. The review delves into the sample preparation techniques, emphasizing the significance of protein precipitation and liquid-liquid extraction in extracting FBS from human blood plasma. LC-MS/MS, chosen for its exceptional sensitivity and specificity, becomes a focal point in FBS analysis, with IS employed to enhance accuracy. The optimization of chromatographic conditions, encompassing the careful selection of stationary and mobile phases, is highlighted as crucial for establishing a robust and reliable LC method, ensuring accuracy in pharmacokinetic studies of FBS. This review positions LC-MS as a cornerstone analytical technique, highlighting its critical role in the precise determination of Febuxostat in blood plasma, with significant implications for future bioavailability and bioequivalence studies. The study concluded that LC-MS's superiority in Febuxostat analysis set the stage for a future where pharmacokinetic studies become insightful, efficient, and transformative, shaping the future of pharmaceutical research.

Phytochemicals, Antioxidant Activity and characterization Studies in the Plant Extracts of Angiopteris evecta (G. Forst.) Hoffm. (Marattiaceae) from Western Ghats | Chapter 5 | Cutting Edge Research in Biology Vol. 7

In the study, phytochemical constituents and antioxidant action of extracts from the Oriental vessel scrappy bushes, Angiopteris evecta (G. Forst.) Hoffm., was examined by various in-vitro procedures. The fronds of A. evecta were collected all along the month of November 2020. The sample was shade drained and grated. The powdered sample was gleaned with ethyl acetate, flammable liquid, and water according to the standard contract. Preliminary phytochemical analysis revealed the closeness of terpenoids, tannins, steroids, reducing sugars, saponins and cardiac glycosides in the extracts. The antioxidant project was evaluated apiece guess of total phenolic content and radical scavenging activity. Folin-Ciocalteau (FC) form was used to determine the total phenolic content present in the extracts of A. evecta utilizing Gallic acid as standard. The methanol extract of leaves written chief total phenolic content of 285.2 µg GAE/g followed apiece aqueous extract (263.87 µg GAE/g). 1,1-diphenyl-2-picrylhydrazyl (DPPH) procedure was employed to measure the radical scavenging potentials. High radical scavenging project was discovered in the ethyl acetate extract (IC50= 88.23 µg/mL).  Further, the leaf extracts was subjected to Thin Layer Chromatography (TLC) and Liquid Chromatography-Mass Spectroscopy (LC-MS) that detected and habitual the presence of saponin parts in the way that Solasurine, Khasianine, Neohecogenin, Alliospiroside C and Maslinic acid. This investigation for the first occasion describes the description of saponin components from the leaf ethyl acetate solid by LC-MS.

A Mini Guide LC-MS and GC-MS Techniques: A Tool for Phytoconstituents Evaluation of Plant Extracts | Book Pi International

Phytochemicals have always been a great matter of scientific investigation because of enormous potential they possess in curing the problems of mankind. Mother Nature is full of plants which have mixtures of phytoconstituents in to each part of them. Such chemicals are obtained by extraction techniques and other processes. However, their purity is the matter of concern. So, scientific world is always in search of such techniques which help in quantitative and qualitative evaluation of the phytoconstituents. Over the years, many analytical techniques have come to the forefront with their specific advantages and limitations. One such thrust area in the field of analytical techniques is chromatography, comprising of simplest one like paper chromatography, thin layer chromatography, gas chromatography to the complex one like high performance liquid chromatography and hyphenated techniques like Liquid Chromatography/Mass Spectroscopy (LC/MS) and Gas chromatography/ Mass spectroscopy (GC / MS). The hyphenated techniques combine the specificity of detection of mass spectroscopy with the resolving power of the liquid and gas chromatography. This technique helps in proper separation of different components. GC-MS technique makes the distinction of structurally similar compounds easier with greater amount of precision with the help of mass spectrometric analysis. The field has its application in separation and purification of the phytoconstituents present in different plant extracts. It is regarded as quantitative measurement method thereby helping in identification of particular class of the phyto-components. Application of LC-MS and GC-MS techniques has helped us to properly characterize such active plant constituents. The chapters elucidated in this book by different authors could help the students and researcher to provide a short review of the techniques stated with plant based examples of different families and different extracts. The team directs each enthusiastic researchers to go in depth to the literature in order to have clear cut understanding of this contemporary area of research.
