Modern Pharmaceutical Technologies for Drug Targeting: A Review on Colon Specific Drug Delivery Systems | Chapter 12 | Challenges and Advances in Pharmaceutical Research Vol. 4

The purpose of the current study is to emphasise the value of contemporary pharmacological techniques for successfully delivering drugs to the targeted site of action by directing them to the colonic area. Due to its potential pharmacological advantages, colon drug targeting has been the subject of much study and development over the past few decades. On the other hand, colon delivery systems have demonstrated their promise as drug carriers for the treatment of many illnesses. They decrease side effects, protect drugs from attacks by the acidic and alkaline medium of the upper GI tract, avoid dosage dumping, etc. The article outlines many technologies or tactics that have been investigated for colon targeting, including time-dependent systems, For prospective pharmaceutical therapeutic applications, consider intestinal pressure-controlled colon delivery capsules, CODESTM technology, pH-dependent systems, enzyme-triggered systems, receptor-mediated systems, etc.

Recent Advancements of In-vitro and In-vivo Evaluation Strategies for Colonic Delivery Systems: An Overview | Chapter 8 | Challenges and Advances in Pharmaceutical Research Vol. 4

The goal of the current review paper is to discuss new developments investigated for colonic delivery system in-vitro and in-vivo assessment methodologies and their prospective pharmaceutical uses. Colon focused medication delivery systems are created in a way that by-passes the gastro intestinal (GI) tract's upper hostile bio-environment while directly releasing material into the colonic area exclusively. This article focuses on in-vitro and in-vivo assessment techniques that can prove a delivery system's resistance to attacks on the upper bio-environment of the GI tract, such as GI motility, pH shifts from an acidic to an alkaline medium, fluid contents, gastro enzymatic activity, etc. These stated assessment metrics support the effective development of medication delivery systems for colon-specific targeting and assist us in overcoming formulation restrictions. The use of a few polysaccharides as excipient materials for the creation of several colon-targeted medications was also a major focus of this research.

A Review on Colon Specific Drug Delivery Systems: Novel Approach in Pharmaceutical Perspective | Chapter 7 | Challenges and Advances in Pharmaceutical Research Vol. 4

The current article's goal is to investigate colon-specific delivery methods and the variables that determine how well drug candidates are delivered to the relevant pharmacological locations. The colon has demonstrated to be a location where medication candidates can be effectively absorbed locally, topically, and systemically. It adopts the preferred oral route for patient compliance and has less systemic adverse effects. This method of medicine delivery is preferable over the painful rectal route, which has lower patient compliance rates. The anatomy and physiology of the colon, colonic pH, colonic flora, GI transit time, fluid quantities, formulation parameters, etc. are some of the variables impacting colon delivery. The use of a colon-specific delivery method for treating disorders like colonic cancer, amebiasis, Crohn's disease, ulcerative colitis, etc. enables drugs to be sent directly to the intended site of action. Direct drug administration into the colon utilising matrix or reservoir-based delivery systems is achievable because these systems can defend themselves against attacks from the stomach's acidic and small intestinal's alkaline environments.
