RP-HPLC Method Development and Validation of Stability Indicating Assay for Simultaneous Determination of Pantoprazole, Diclofenac, Chlorzoxazone in Pharmaceutical Dosage Form | Chapter 6 | Pharmaceutical Science: New Insights and Developments Vol. 1

Background: High-performance liquid chromatography (HPLC) is the fastest-growing analytical technique for the analysis of drugs. Its simplicity, high specificity and wide range of sensitivity make it ideal for the analysis of many drugs in both dosage forms and biological fluids.

Objective: A simple, accurate, precise method was developed, for the simultaneous estimation of the Chlorzoxazone, Pantoprazole, and Diclofenac in solid dosage form.

Method: The chemicals used were Water-HPLC grade, Acetonitrile, Triethylamine, Potassium dihydrogen orthophosphate, and Orthophosphoric acid. Chromatogram was run through BDS C18150×4.6mm, 5m. Mobile phase containing 0.01 % KH2PO4 and Acetonitrile in the ratio of 52:48 v/v was pumped through column at a flow rate of 1.0ml/min. The buffer used in this method was 0.01% KH2PO4, Temperature was maintained at 30°C. The optimized wavelength for Phar Chlorzoxazone, Pantoprazole, and Diclofenac was 229.0 nm.

Results: In the estimation of the three drugs, four trials were conducted using various solvent systems, columns, and experimental conditions. The retention time of Chlorzoxazone, Pantoprazole, and Diclofenac were found to be 2.229 min, 2.958 min, and 3.568 min. The percent RSD of system precision for Chlorzoxazone, Pantoprazole and Diclofenac were 0.9,0.6 And 0.6, respectively. The percent RSD of the method, Pantoprazole and Diclofenac were found to be 0.6, 1.0 and 0.7 respectively. The percent recovery was obtained as 99.86%, 100.07% and 99.70% for Chlorzoxazone, Pantoprazole, and Diclofenac respectively. LOD values obtained from regression equations of Chlorzoxazone, Pantoprazole and Diclofenac were 0.14 ppm, 0.24 ppm, and 1.83 pm, and LOQ values obtained from regression equations of Chlorzoxazone, Pantoprazole and Diclofenac were 0.42 ppm, 0.72 pm, 5.53 ppm respectively. The regression equation of Chlorzoxazone was y = 8321.9.x + 1397.8 Pantoprazole was y = 9806.1x +6071.7 and of Diclofenac was y = 2575x + 4338.4.

Conclusion: Sensitive detectors have transformed liquid column chromatography into high speed, efficient, accurate and highly resolved methods of separation. Retention times were decreased so that the method developed was simple and economical hence it can be used in regular Quality control tests in industries.

A Histopathological Study of Proton Pump Inhibitors: Are They Safe on Kidneys? | Chapter 14 | Technological Innovation in Pharmaceutical Research Vol. 9

Background: PPI is used in between 45 and 85 percent of instances in India. In 82.96 percent of cases, patients with chronic renal illness were prescribed a PPI. Recent case reports, however, suggest that PPI can cause AIN. Because many patients take many drugs, it's tough to estimate the exact rate of this uncommon side effect, making establishing a causal relationship between AIN and PPI much more challenging. Early detection of AIN and withdrawal of the offending medicine may delay the progression of AIN to end-stage chronic renal disease. As a result, we decided to conduct research to see if there is a link between PPI and AIN. The goal of the study was to see what effect concomitant usage of PPI and NSAID or AMA had on the histopathology of experimental animals. The goal was to use histopathological investigations to determine the effect of simultaneous treatment of PPI with NSAIDS/antimicrobials on kidneys and compare it to a normal histological picture to see what changes had occurred due to medication administration. The rats used in the experiment weighed between 150 and 250 grammes. Omeprazole, Three groups were given pantoprazole, rabeprazole, and diclofenac for 28 days: group A received nothing, group B received diclofenac, and group C received ofloxacin. The animals with disturbed RFTs were grouped together after the 28th day. Two animals were sacrificed for each group. Their histopathology tests were also finished. The RFTs of at least three rats in each group were found to be aberrant. And, whether or not AIN is used, the majority of histopathological studies show structural and vascular changes. Conclusions: PPIs alone are known to cause AIN, but when other nephrotoxic medications are taken, the risk of AIN increases.

A Randomised Study to Compare the Efficacy and Tolerability of Aceclofenac and Diclofenac in Osteoarthritis of Knee Joint | Chapter 10 | Highlights on Medicine and Medical Science Vol. 3

Background: Osteoarthritis is a slowly progressive condition characterized by hyaline cartilage loss. Nonsteroidal anti-inflammatory drugs are important in the treatment of osteoarthritis. Objectives: The purpose of the study was to compare the efficacy and tolerability of Aceclofenac and Diclofenac in patients with knee osteoarthritis. Material and Methods: This is a double-blind, randomized parallel group study. The study included 60 patients with confirmed knee osteoarthritis; 30 were given Aceclofenac and 30 were given Diclofenac after food for 8 weeks, and the patients were evaluated and compared. Pain intensity by VAS Score, joint tenderness, swelling, erythema, pain on movement, functional capacity, and overall assessment on the LIKERT Scale were all statistically significant. Both drugs resulted in significant improvements in the parameters of pain intensity, joint tenderness, swelling, erythema, and pain on movement of the OA knee joint, but aceclofenac resulted in greater improvement in all parameters. Conclusion: Aceclofenac is the preferred NSAID for osteoarthritis of the knee joint over Diclofenac because it is more effective. However, there was no significant difference in the adverse effect drug profile.
